This system was used to analyzed in vitro cultures of differentiated pluripotent stem cells. The Evos FL allowed to simply analyze fluorescent signals from cell culture and histological stainings at various magnification. The system has a small fooprint and does not require a dark room.

Protocol Overview

Cells were cultured and differentiated on various dishes ( p35, p60) and multi-well plates (96, 24, 6 well-) format, followed by fixation and immunohistochemistry directly on culture plates. Vessel was placed on the Evos FL stage with the specific holder and desired magnification (10-20X) was selected manually. Fluorescent channel (up to four) was also selected manually and exposure parameters were defined from the digital display from optimal image generation.

Tips

Change stained cultures to PBS saline to lower autofluorescent background. For co-staining, choose fluorescent channels far away along the color spectrum.

Results Summary

Good resolution (up to 4MB) B&W Tiff pictures were generated for each fluorescent channels from live cultures (also worked on various live cultures, expressing fluorescent molecules) ranging from 4X to 40X. Scale bar could optionally be added. Images were directly saved on a USB key plugged on the system, in dedicated folders.

Excellent system that can be use routinely for fluorescent stainings and/or signals acquisition. Good selection of filter cubes for standard dyes/colors. Compatible to various culture/sample format., small footprint, does not require a dedicated (dark) room.
