Generally, we don't see to much background caused by biotin/streptavidin except for specific tissue samples, so we never had a need for a blocking kit for those. We recently took on a project that has a lot of background sources (mouse-on-mouse, biotin/streptavidin, peroxidase), so it was necessary to look for a blocking kit. In addition, we were asked by an investigator to replicate results from a previous protocol using a similar blocking kit. We would normally prefer to use polymer-based detection methods (such as with a Leica Bond RX), but we went with ABC-based method to match the previous protocol. We went with Vector Labs because we get a lot of our biotinylated secondaries and other associated reagents from them and have always had great results. The same was true with SP-2002. It was R.T.U. and required no additional tweaks. Just slide the steps in right before your primary antibody.
