The goal of this research was to clearly label mouse CD19+ B cells for phenotypic analysis of B cells in different bodily tissues, including the spleen, lymph nodes, and peritoneum, by flow cytometry. This product was selected to compare binding density versus another popular monoclonal CD19 mAb clone.

Primary Incubation

Following splenocyte isolation by gentle dissection and red blood cell lysis, wash cells once in FACS buffer (PBS plus 1% BSA or serum). Block Fc receptors and stain for viability in PBS for 20 minutes on ice.

Secondary Incubation

Wash cells once in FACS buffer. Stain with isotype control or CD19 antibody (1:500 dilution, 100 uL per million cells) for 20 minutes on ice.

The Good

Clearly labels CD19+ B cells.

There are brighter antibodies directed against CD19.

The Bottom Line

Because this antibody isn't as bright as some other available CD19 antibodies, it can be useful in instances where a lot of other channels are being engaged, so the downstream compensation will not be as significant. However, if one needs to robustly distinguish B cells from non-B cells to remove potentially confounding populations, a brighter antibody may be more useful.
