Our laboratory investigates different ways to improve immune responses against HIV, including enhancement of the capacity of NK cells to kill HIV-infected cell. We are currently investigating the capacity of an engineered antibody to trigger NK-cell mediated ADCC activity in in vitro assays against an HIV-infected cell line. We perform FACS-based killing assays, where in addition to target cells killing we look at degranulation of NK cells and release of granzymeB and perforin. Good and strong fix/perm buffers are needed to stain for these factors intracellularly, so we use this kit.
Intracellular staining for FACS
Whole human PBMC, or purified NK cells from human PBMC
We follow the protocol recommended with the kit, and dilute the buffers to their suggested ratios.

We get great and clean staining using this kit. We have use other kits from other companies in the past, but this one yet remains the best in our hands.
