HEK293 Whole Cell Lysate

Protocol Overview

Cells were first transfected with plasmid (Luciferase Reporter Vector ) of interest along with Renilla luciferase vector. After 36 hours of transfection, cells lysed with 50 microlitre (per well/24 well plate) of 1 X Passive Llsis buffer. 10 microlitre of each sample has taken in 1.5 ml centrifuge tube and 50 microlitre of Luciferase Assay Reagent II added to generate luminescent signal, which was measured by luminometry. Immediately, 50 microlitre of Stop & Glo reagent was added to the same tube to quench the reaction and initiate the Renilla luciferase reaction, which was also measured by Luminometry. The relative luciferase activity was measured by normalizing with Renilla luciferase activity.

50 microlitre of Luciferase Assay Reagent and 50 microlitre of STOP & Glo is enough for measurement

Results Summary

The kit produces high counts of luminometry measurements of both firefly luciferase and renila lunicferase activity.
