I used this kit to isolate human CD4+ T-cells from PBMCs. This bead based column isolation, helps me to use both positive and negative selection fractions for analysis.

All that you need to do is just follow the protocol from Miltenyi as such. Always count the initial number of cells taken for isolation. Don't vary with antibody cock tail to be used and number of washes recommended as it may affect final purity.
Protocol is very straight forward and simple and just follow it.
I always get more than 90% purity.
