I used this buffer for fixing/permeabilizing cells to analyze Foxp3 transcription factor expression by flow cytometry. General fix/permeabilizing buffers will not work for nuclear staining. So I selected this particular buffer set.

Mix 4x fix/permeabilizing buffer with 1x dilution buffer. add to washed cells at 50 micro liter per 100 -200 thousand cells. Incubate at 4 degree for 1h.
Works well for signaling kinase staining assys as well. Use 1X perrm/ wash buffer (Tonbo biosciences) for subsequent washes.
After incubation washed the cells with perm/wash buffer and continued normal staining procedure. Worked fantastically for Foxp3 staining even after a week of incubation.
