The goal of this experiment is to determine if protein kinase R is activated upon virus infection in primary mouse macrophages. To address question, the levels of eIF2a and p-eIF2a were examined by western blotting using specific antibodies (Abs) against eIF2a and p-eIF2a. The Ab against mouse eIF2a is a mouse monoclonal Ab (sc-133132) purchased from Santa Cruz. The western blot results shown that this Ab has a high specificity.

1:200 dilution in TBST; 4 degree overnight incubation

Blocking Agent

5% non-fat milk in TBST at RT for 1 hour

1: 10000 dilution in TBST with goat anti mouse IgG at RT for 1 hour

Membranes were washed and visualized for chemiluminescence in Western Lightning Plus-ECL solution

Results Summary

Mouse eIF2a was show in a single band on the membrane without non-specific bands, indicating a high specificity of this Ab. Calnexin was used as loading control.
