I looked for CD11b expression at the cell surface of live HL-60 cells by flow cytometry. This product comes with a commonly used fluor that requires very little optimization to use.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

HL-60 (Human promyelocytic leukemia cells) cell line

Primary Incubation

2.5 ul antibody in 200 ul PBS for 1 h at 37 degrees C

Blocking Agent

N/A

Secondary Incubation

N/A

Tertiary Incubation

Detection

Flow Cytometry

Results Summary

HL-60 cells were treated with all-trans retinoic acid for 48 h and live cells were stained with the anti-CD11b antibody. Expression was analyzed via flow cytometry. Retinoic acid treatment induces expression of CD11b at the cell surface, and we were able to detect significant expression of the protein.

Summary

The Good

Relatively inexpensive, does not require optimization, reproducible results

The Bad

Peaks often have a long tail not always present using other CD11b antibodies

The Bottom Line

All factors considered, this is a good, effective antibody. It is consistent and relatively inexpensive, requiring little or no optimization.
