In the present study retinas were extracted from artificially induced type-2 diabetic and non diabetic rats. For this, rats were sacrificed by following standard ethical guidelines, eyes were enucleated, retinas were isolated and protein extracts were prepared in RIPA buffer. Following, equal amount of proteins were resolved in 10% SDS PAGE and immunoblotted to check pYAP levels. Antibody was selected on the basis of information’s provided in antibody flyers.

Overnight at 4 0C (5% BSA in TBS with 0.1% Tween 20; dilution 1:750)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 2 hr at RT)
1 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:4000)

pYAP antibody from cell signaling technologies has given specific but slightly faint band at desired molecular size. Some non specific proteins were also observed at 45 kDa in rat protein.
