Present study was performed to test the effect of specific nutritional treatment on SIRT1 levels in liver of diabetic male wister albino rats. In brief, after specific treatment rats were sacrificed at different time points, liver were harvested, protein extract were prepared and resolved in SDS-PAGE. Resolved proteins were subjected to immunoblotting for SIRT1 proteins. Antibody was selected from abcam because of its merit indicated in antibody flyer.

Overnight at 4 0C (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:1500)
5 % Skimmed milk in TBS with 0.1% Tween 20 (for 2 hr at RT)
2 hr at RT (5% skimmed milk in TBS with 0.1% Tween 20; dilution 1:4000)

Very bright and intense band of SIRT1 at 150 KDa were observed. There were no or very little non-specific bands.
