We need high amounts of plasmid DNA to make lentiviruses to use in our research on TCRs. We selected this product because it is easily available at our facility's freezer program and we have had good results from other invitrogen's kits in the past.

Plasmid DNA isolation from bacterial culture

Starting Material

Bacterial culture (Stbl3 competent cells)

Protocol Overview

Spin down an overnight bacterial culture that was transformed with the plasmid of interest. Use the buffers from the kit to lyse the cells, precipitate, neutralize and apply the culture to the maxiprep column. Wash the column. Elute. Add isopropanol and spin for 30min at 4C. Wash the pellet with 70% ethanol, dry and solubilize the DNA.

Tips

You can use 500ml (instead of 250ml) of bacterial culture for one batch to maximize the DNA isolated. Make sure the isopropanol and 70% Ethanol have been chilled on ice. I find that this also increases DNA yield.

Results Summary

I consistently get up to 1.5mg of DNA from a single maxiprep.
