We are interested in enhancing the killing capacity of HIV-specific CD8 T cells by reengineering TCRs expressed by these cells. We perform in vitro killing assays where CTLs activity is tested in presence of HIV-1 infected cells. Human T cells are purified from donors to isolate CD8 T cells, the effector cells, and CD4 T cells, the HIV-infectable target cells. We use human CD3 antibodies to study the purification quality, as well as to detect the cells in FACs assays.
