Our lab works on an antibody with enhanced capacity to mediate ADCC against HIV-infected cells. We set up in vitro assays with purified NK cells and HIV-infected target cells to test the capacity of the antibody to mediate clearance of infected cells by NK cells. Given that CD16 (FcgammaRIIIa) is the primary receptor expressed on NK cells that binds to the anti-HIV antibody, it is crucial for us to detect the expression levels of the receptor in order to interpret the assay results correctly.
