Protocol Overview

PBMCs are purified from whole blood, counted and incubated with the beads needed to separate (or deplete) a specific cell population. After 30min incubation with beads the cells are resuspended in running buffer and set up in the machine which does the rest of the work.

Tips

I always separate the sample in 5 15ml conical tubes (no matter the starting cell number). I get a better yield that way.

Results Summary

The machine works great when it works. We had many problems with it and the Miltenyi reps always came to fix whatever was wrong as long as the machine was still under the service contract/warranty. After that they wanted to charge us up to $8000 for a single visit to come and fix the machine. We eventually stopped using it due to the high maintenance cost. The manual separation is more time consuming but overall slightly faster.

Features Summary

Many technical problems. With frequent use we needed someone to come out and fix small things every few months.
