The Direct-zol kit merges a miniprep format with Trizol RNA isolation. The benefit of the kit is that is significantly reduces the time needed to purify RNA samples when lysing cells or tissues with Trizol, RNAzol, or any other TRI reagent.

A cell scraper is very handy when lysing monolayers from a tissue culture plate.

Results Summary

As can be seen in Figure 1, RNA bands are distinct with little gDNA contamination. This experiment yielded about 200 ng/uL of RNA (about 10 ug total) from a single well of a 12 well tissue culture plate. In Figure 2, four separate RT-PCR reactions were performed using the purified RNA. The BTF3 product was the best yield; DRG1 less so [note arrow]. The other two reactions require optimization.

Additional Notes

For RT-PCR, I strongly recommend the in-column DNAase treatment.
