The aim of this project is to study different types of T-cells in human colon cancer. In this particular experiment, human colon cancer tissue microarray is stained with CD8 antibody. I had purchased this antibody with no prior knowledge on whether this performs well for this specific purpose.

Primary Incubation

1:100 dilution in blocking buffer; over night incubation at 4 deg Celsius

Blocking Agent

1xPBS + 1% BSA + 1% goat serum

Secondary Incubation

1:200 dilution (in 1xPBS + 1% BSA) of Goat-anti mouse Alexa dye 488; 1 hour at r/t0 dilution (in 1xPBS + 1% BSA) of Goat-anti rabbit Alexa dye 594; 1 hour at r/t

Tertiary Incubation

DAPI included in the last wash step

Results Summary

CD8 T-cells were clearly detected in the human colon cancer tissue microaarray core biopsy sections.
