This antibody was used in a study investigating the myopathic effects of stains using C2C12 myotubes. It has been published in Biochimica et Biophysica Acta in 2015 (Chu et al., Endothelial protective genes induced by statin are mimicked by ERK5 activation as triggered by a drug combination of FTI-277 and GGTI-298, Biochimica et Biophysica Acta. 1850, 1415-25). Anti-ERK5 antibody was used to confirm ERK5 knockdown after siERK5 transfection in HUVEC cells.

Western Blot

Sample

Total protein lysate from HUVEC cells 24 hours post-transfection with siRNA against ERK5.

Primary Incubation

1:1000 in 1% milk incubated at 4C, overnight.

Blocking Agent

5% milk.

Secondary Incubation

Anti-Rabbit-HRP (1:3000) in 2% milk incubated for 1 hour at room temperature.

Tertiary Incubation

N/A

Detection

SuperSignal West Femto ECL Substrate (Thermo #34094).

Results Summary

Anti-ERK5 worked great to detect endogenouse ERK5 protein in HUVEC total cell lysate.
