I use this transfection kit for making lentivirus making on the 293T cells. I usualy get 70-80% transfection efficiency with this kit.
Highly efficient transfection
293T cells, plasmid /library DNA
The day before transfection 293T cells plated on the 10cm dish with 0.5million cells. On the day of transfection the cells density was close to 95%, I used to 10ug of lentiviral plasmid carrying mcherry tag, 30ul of TransIT293 reagent. I can observed mcherry expression after 24hr on the cells. I never observed any toxic effects on the cells.
Maintain plasmid: transfection reagent ratio 1:3
After 48hr nearly 80-90% cells were positive for mcherry expression.
