Our lab uses the CD4 T cell isolation kit to obtain cells for in vitro and in vivo mouse antigen specific proliferation assays. We have been able to successfully and repeatedly obtain pure (90%) and viable (90%) T cells with this negative isolation kit, however the time to run the entire protocol is long. On average it takes two hours which can be harsh on primary T cells.

Make sure experiments and components are kept cold, since it takes so long to complete
We obtained viable and pure (90%) CD4+ T cells after the isolation process which were later used to generate antigen specific proliferation assays. Cells were able to proliferate (99%) in response to an antigen.
