Cloning of the full length desired gene requires amplification of the gene of interest and extraction of the amplified gene from the agarose gel. Invitrogen pure link quick gel extraction can be used for extraction of the desired fragment from the agarose gel after its separation. High yield of DNA can be recovered from the gel by using this kit  which can be used directly for ligation process.

Protocol Overview

The desired DNA fragment is excised from the agarose after eletrophoritic separation and weighed . 3 X volumes of the gel stabilization buffer is used to solubilize the gel loaded on to the column and spun at 12000g. The column is washed with wash buffer and finally eluted using elution buffer

Tips

Do not take more than 300 mg of gel

Results Summary

A good quantity of DNA is recovered from the gel which can be used for sequencing or cloning in to a desired vector
