This kit was used for quantifying TNFa in plasma samples of chickens for immune comparative analysis. We selected this kit by appointment of another researcher.
Quantifying TNFa in body fluids of chickens
21 days chicken plasma
1) prepare reagents and samples;2) set a blank well without any solution;3) Add 50uL of samples or standards in each well;4) add 50uL of HRP-antibody in each well (except in Blank well);5) Incubate for 40 min at 37ºC;6) Wash 5 times;7) Add 90uL of TMB soluiton and incubate for 20 minutes at 37ºC);8) Add 50uL of Stop Solution and read the absorbance at 450nm;9) Calculate the TNF concentrations.
Pay attention: this is a competitive ELISA, so greater the absorbance, lower the concentration of TNFa in samples.
The curve os standards is very weird, the detection range (0,27 to 200 pg/mL) seems random.The curve was drawn on a logarithmic scale, and the equation for quantyfing was obtained with a linear regression. The r² was iqual to 0,9863.
