I routinely purify monocytes for differentiation into monocyte-derived DC and macrophages for testing responses to an immunomodulatory therapy. I selected this product in order to obtain untouched monocytes from PBMC after density gradient centrifugation.

The protocol, in brief, consists of resuspending isolated PBMC in a PBS buffer, followed by incubation with the negative selection antibody cocktail. Next, magnetic beads are added for an additional brief incubation. Cells are purified by placement in a magnet and removal of the supernatant containing monocytes.
Vortex the magnetic beads thoroughly to avoid clumping
Monocyte purities with this protocol are routinely 75% or greater and high viability.
