Good Ab for flow. We study inflammation during acute Mtb infection, and as a result, our timepoints look at primary cells from lungs from extremely activated conditions, in which we find CD3 or TCRb staining always to be lackluster. We've done some CD3 staining with vaccinated mice or uninfected and find it to be much clearer than during Mtb infection, so I think the unimpressive staining we see is a result of our cells, not because of the quality of Ab.That being said, we have found that TCRb staining looks better than CD3, and will probably use that in the future for general T cell gating. This stain was used mixed with all our normal Ab (surface) mixes, and also worked fine with EdU staining (PFA fixation, saponin perm and ClickIT reaction in the presence of Copper containing buffer). Worked fine with FITC - which is nice and much cheaper than 488 conjugates (I prefer 488 for other things, but the FITC was sufficient for our purposes).

Nice staining for TCRb, good pre-gate used to identify T cells, works well for our purposes. I like it better than CD3 and the cheaper FITC conjugate is sufficient for our experiments. (Although I haven't been doing any detailed T cell analyses, just basic CD4 and CD8 counts) Keep in mind that TCR in down regulated / internalized during activation conditions so its not always the prettiest stain depending on the sample. Many product sheets use an unstimulated population for their staining, and so it looks a lot better there for that reason.
