The rabbit antibody bs-10257R from Bioss USA was used for immunostaining of COX4 in human monocyte-derived macrophages permeabilized with Triton X-100. The antibody produced acceptable staining consistent with mitochondrial localization of COX4.
Immunofluorescence
Human monocyte-derived macrophages (grown on glass slides, fixed in 1% formaldehyde in PBS, permeabilized with Triton X-100)
Overnight, at 4-8 degrees Celsius, at 1:200 dilution
10% normal donkey serum; 1 hr; RT; no permeabilization
Donkey anti-rabbit IgG conjugated with DyLight649 from Biolegend, 1:500, 1 hr
NuncBlue was used to highlight cell nuclei
IF microscope; Olympus
The antibody produced acceptable results being used at a relatively low final concentration of 0.5 ug/mL. The staining was consistent with the mitochondrial localization of COX4.
