Primary Incubation

Overnight, at 4-8 degrees Celsius, at 2 ug/mL in PBS/1% BSA

Blocking Agent

10% normal donkey serum; 1 hr; RT

Secondary Incubation

Donkey anti-rabbit IgG conjugated with DyLight649 from Biolegend, 1:500, 1 hr

Tertiary Incubation

NuncBlue was used to highlight cell nuclei

Detection

IF microscope; Olympus

Results Summary

The anti-COX4 rabbit antibody A301-899A from Bethyl produced good staining in human monocyte-derived macrophages permeabilized with saponin. The staining was consistent with the mitochondrial localization of COX4.
