Rat monoclonal IgG2a antibody against mouse/human Mac-2 (Galectin-3) (Clone M3/38) from Cedarlane used for immunofluorescent staining of human monocyte-derived macrophages. This antibody produced excellent results with non-permeabilized cells, confirming the plasma cell localization of Mac-2/galectin-3 protein.

Human monocyte-derived macrophages (grown on glass slides, fixed in 1% formaldehyde in PBS, no permeabilization)

Primary Incubation

Overnight, at 4-8 degrees Celsius, at 10 ug/mL in PBS/1% BSA

Blocking Agent

10% normal goat serum; 1 hr; RT; no permeabilization

Secondary Incubation

Goat anti-Rat DyLight488 antibody from Biolegend (used at 1:500, 1 hr, RT)

Tertiary Incubation

NuncBlue was used to highlight cell nuclei

Detection

IF microscope; Olympus

Results Summary

The antibody CL8942AP against Mac2/galectin-3 produced excellent staining of human non-permeabilized monocyte-derived macrophages. I would highly recommend this antibody for immunostaining of Mac2/galectin-3-positive cells.
