The rabbit polyclonal antibody sc-894 from Santa Cruz was used for immunofluorescent detection of caveolin in human prostate cancer cells. In formaldehyde-fixed cells, this antibody produced excellent staining at 1:500 dilution.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Human prostate cancer PC-3 cells, grown on glass coverslips, fixed in 1% formaldehyde for 20 min RT, and permeabilized in 0.2% Triton X-100 for 5 min, RT.

Primary Incubation

Overnight, at 4-8 degrees Celsius, at 1:500 dilution (0.2 ug/ml)

Blocking Agent

5% donkey serum in PBS 0.001% Triton X-100

Secondary Incubation

Donkey anti-rabbit IgG conjugated with DyLight488 from Biolegend, 1:500; 1 hr

Tertiary Incubation

NuncBlue was used to highlight cell nuclei

Detection

IF microscope; Olympus

Results Summary

The antibody produced excellent staining in formaldehyde-fixed and permeabilized prostate cancer cells.

Additional Notes

This antibody worked extremely well also in human monocyte-derived macrophages.
