We routinely in our lab purify and concentrate lenti virus using this kit. The procedure is very simple and takes 30-40 min per sample. We can concentrate up to maximum 30ml lenti viral supernatant per column. The protocol does not need any high-speed centrifuge and quality of virus is good with our hands.

The protocol is very simple. The viral supernatant was mixed with 10X dilution buffer after removing the cell debris from the viral harvest. The viral -dilution buffer mix passed through column (supplied with kit) which might take 10 min per column. Then column was washed with wash buffer 20ml and then eluted with 3ml of elution buffer. The eluted was frozen immediately for downstream application.
Don"t dry the column
The lenti virus was free contaminant from medium which might hinder the infection efficiency. Final viral is concentrated upto 10 fold after the purification.
