The anti-human E-cadherin (CD324) antibody # 324101 from Biolegend is advertised as capable of detecting E-cadherin on the surface of human cancer cells by a variety of methods, including flow cytometry, immunostaining and western blotting. In our hands, however, this antibody failed to detect E-cadherin by both flow cytometry and cell immunostaining in human prostate cancer cells known to express high levels of E-cadherin. Therefore, I would not recommend this antibody for detection of E-cadherin in human tumor cells either by flow cytometry or immunostaining.

The antibody #324101 from Biolegend did not recognize any cell surface E-cadherin (i.e. in the cells without permeabilization), in contrast to the anti E-cadherin mAb from Becton Dickinson.

I would not recommend this antibody for detection of cell surface E-cadherin on human cancer cells.
