Company:

Abcam

Product Name:

Anti-MAP2 Antibody

Catalog Number:

Ab32454

We needed a good MAP2 antibody to assess the immunoreactivity of this neuronal marker in human retinoblastoma cells. We chose this particular antibody because it had a wide range of immunoreactivity (mouse, rat, goat, cat, human) and had an excellent rating on the Abcam website.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Fixed cytospins of Y79 human retinoblastoma cells

Primary Incubation

MAP2 primary antibody (10 ug/ml) was incubated for one hour at room temperature in PBS with 0.05% Tween-20

Blocking Agent

None

Secondary Incubation

45 minutes at room temperature with rabbit anti-Dylight 488 (Vector D11488) at 5ug/ml in PBS with 0.05% Tween-20

Tertiary Incubation

Vectashield mounting medium containing DAPI counterstain

Detection

Fluorescence microscopy- Nikon microscope with SPOT camera

Results Summary

The MAP2 antibody provided excellent results as seen in the accompanying figure. We observed a low-to-high range of immunofluorescence on a cell-by-cell basis, which is what we would expect from a heterogeneous neuronal tumor.

Summary

The Good

Easy to optimize with low background

The Bad

No downside

The Bottom Line

This antibody works very well for immunofluorescent staining of human cells and would be compatible with other primary antibodies (mouse, rat, chicken, etc.) for double immunostaining experiments.
