This Kit Did Not Work.

The manual contains many mistakes and unclear instructions. However, I communicated with tech support at Abcam, and got more clear instructions. I then followed exactly what they said to do. I attempted to use the kit several times with my cells (adherent mammalian cells in cell culture plates) doing it the way tech support said to do. The kit claims to measure total NADP+NADPH, and also NADPH, and also NADP. Each time I used the kit, I got wildly unpredictable results. For example, the total NADP+NADPH would be much more than the sum of NADPH and NADP measured independently. Or, the NADPH alone, or the NADP alone, or both of them, would individually be much more than the so-called total. I contacted Abcam tech support many times about this and they were very nice, and one of the tech support team actually came over to our lab (we're in Boston) and sat with me at my computer going over my spreadsheets for a couple hours one day to see if there was anywhere I might be making a mistake or anything. Finally, they told me to try a positive control that had always worked in their hands: HeLa cells. So, I grew some HeLa cells and tried them instead of my regular cells. I got bad results (NADPH alone measured much higher than total NADPH + NADP) even with authentic HeLa cells. I spent several weeks total working on this, Abcam sent me a replacement kit from a different lot for free (which was very nice of them), and still I could not get reliable results. Also, the standard curve was always very curved when the manual says it is supposed to be straight. After all this effort, I still do not know how much NADP and NADPH are in my cells. So Abcam is giving me my money back. This is very nice of them, and I appreciate that. They are a great company to work with, but the kit is not any good.
