Application

Concentrating samples

Starting Material

Peptides in Formic acid/acetonitrile

Protocol Overview

For over 10 years we have been concentrating samples using an old 3 way setup: a ventilated/heated centrifuge, a cold-trap and a pump. It was circumlocutory to use and requires a lot of space. The Eppendorf concentrator plus is a benchtop vacuumcentrifuge that is very easy to use and requires almost no setup. A timer can be set for the time required to dry and it will stop spinning after the time has passed, it will continue to heat though. It has 4 temperature settings: the (current) RT, 30°C, 45°C and 60°C. The brake can set to on/off. It also has several different functions which regard the vacuum to be applied. The strongest vacuum will be V-AQ(aqua solution) followed by V-AL(alcoholics), and V-HV(high vapor), these 3 functions will spin the rotor and turn on the vacuum. The concentrator plus can also act as a desiccator with the same settings without centrifuging. Finally it can also be used as a normal centrifuge, however the speed cannot be set.

Tips

Do not fill plates for more than 2/3 of the volume as stated by Eppendorf.

Results Summary

It takes roughly 1h and 45min to concentrate 48 eppendorf tubes from ~500µl to dry.

Features Summary

Extremely easy to use and foolproof.

Additional Notes

Be careful with overfilling plates.

Summary

The Good

Easy to use, foolproof, benchtop(size).

The Bad

Only 4 temperature settings, speed cannot be regulated, plates can only be used for 2/3 of the volume.

The Bottom Line

It's a great benchtop concentrator (vacuumcentrifuge). Easy to setup out of the box (5 mins) and works like a charm. Plates could be a bit better though.
