Got a free kit from NEB in a vendor show and decided to give it a try. The QIAGEN mini prep kit is used as a control to evaluate this Monarch mini prep kit.

1.5 mL of bacterial culture (DH5 alpha with pENTR-D-TOPO plasmid)

Protocol Overview

The protocol is similar to many other miniprep kits. First, pellet the bacterial culture and resuspend it in Buffer 1 (B1). Then lysis buffer (B2) is added to the tube and invert the tube 5-6 times. Then add the neutralization buffer (B3). Wait for 2 min before centrifuge 2-5 min. The supernatant is then transferred to the collection tube. The rest will be just spin and wash followed by addition of elution buffer.

Tips

Make sure the bacterial pellet is fully resuspended

Results Summary

The purified plasmid DNA is measured by nanodrop from Thermofisher. The quality of purified DNA between NEB and QIAGEN kit is same. However, the NEB Mornach kit recovers less amount of plasmid DNA

Overall, this is a good miniprep kit and it is as good as Qiagen, except the yield is somewhat lower.
