This antibody was used to identify CD3+ T cells in human peripheral blood mononuclear cells (PBMC) by flow cytometry. This antibody gave excellent results in FACS staining with clear separation of CD3-positive and -negative cells.

Primary Incubation

FITC anti-human CD3 antibody (BioLegend 300306), incubated with approximately 1x10^6 cells for 1 hour on ice at 1:20 dilution in a total volume of 100ul PBS+2% FCS, 1mM EDTA, 0.1% sodium azide.

Results Summary

This antibody worked well in flow cytometric analysis of human PBMC. CD3 staining was bright and cells were well separated from negative cells, allowing easy identification of CD3-positive lymphocytes. Only minimal background staining compared to unlabelled cells was observed; however, some users may want titrate this antibody further so that negative cells exhibit identical fluorescence to unlabelled cells.

Additional Notes

For flow cytometric analysis, gating was performed on the lymphocyte population. Monocytes were negative (not shown).

Summary

The Good

CD3 staining was bright allowing easy identification of CD3-positive lymphocytes. Extracellular epitope, thus fixing and permeabilization of cell is not required. Excellent pricing. Directly conjugated with a fluorophore resulting in simple cell staining protocol. Suitable for use with additional techniques (reported by the manufacturer).

The Bad

The Bottom Line

This is a very good antibody to detect surface expression of CD3 on human PBMC. Bright staining of positive cells allowed easy quick and easy discrimination between positive and negative cells. Furthermore, the fact that this antibody is directly conjugated to FITC negates the use of a secondary antibody which reduces the time required for cell staining. Highly recommended.
