To study the mechanisms of endothelial-to-mesenchymal transition. This particular experiment was to detect the expression of HDAC7 in human coronary artery endothelial cells before and after knockdown with shRNA. HDAC7 shRNA and antibody were both purchased from Santa Cruz Biotech.

Lysed Human Coronary Artery Endothelial Cells

Primary Incubation

1:500 dilution in 5% skim milk incubated at 4 degrees celcius overnight

Blocking Agent

5% skim milk for 1 hour at room temperature

Secondary Incubation

5% skim milk with Goat anti-mouse IgG conjugated to HRP. 1:15,000 dilution for 2 hours at room temperature

Tertiary Incubation

Detection

Enhanced chemiluminescence for 2 minutes

Results Summary

Clear bands at expected molecular weight of 105kDa. Lane 1/2 is scrambled shRNA control, lane 3/4 shows knockdown of HDAC7 protein using HDAC7 shRNA. 25ugs of protein loaded into each well after 5 minutes of boiling in laemmli buffer. Imaged on a Biorad western blot imager.
