Company:

Abcam

Product Name:

Anti-NQO1 [clone A180]

Catalog Number:

ab28947

We evaluated relative expression of NQO1 across a panel of ovarian cancer cell lines. I chose this antibody because I typically have success with Abcam antibodies by Western, and the price was reasonable.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Ovarian cancer cell line panel - lysates

Primary Incubation

1:2000 in 5% milk in TBS-T; incubated overnight at 4 degrees C on a rocker

Blocking Agent

5% milk in TBS-T

Secondary Incubation

1:5000 anti-mouse HRP in 5% milk in TBS-T; incubated 30 minutes at room temp on a rocker

Tertiary Incubation

NA

Detection

HRP Chemiluminescence

Results Summary

There was high, unexplainable background on the blot. Even after the assay was repeated 3 times, there remained high background. The antibody was used initially at 1:1000, then at 1:2000 and 1:2500, but the background remained at 1:2000 and the bands were far less detectable at 1:2500.
