We are mainly working on obese retinal tissue in the lab. During my research I check the many protein expression pattern in retinal tissue. For this I use immunofluroence technique. For the above reason I use the ABC kit from Vector Laboratories.
Protein level expression in retinal scetions
Control and obese rat retinal paraffin sections.
Retinal paraffin sections, deparaffinized with xylenes and hydrate through graded series of alcohols. Ag retrieval step was done with citrate buffer. Blocking is done with goat serum for 1 hr. Afterwards, wash incubated with primary Ab overnight at 4oC. Then washed and added secondary antibody of respective primary. Finally, developed with kit.
Prepared all the reagents according to manufacturer's guidelines.
Tried with GFAP in retina of obese rat, it worked well.
