Company:

Thermo Fisher Scientific

Product Name:

Rhodopsin Antibody (1D4)

Catalog Number:

MA1-722

Rhodopsin, is a G-protein family which is present in the photoreceptor layer where rods and cone cells are present. This protein detect the degree of lightness entering the retina and their sensitivity is dependent on the amount of Rhodopsin present which is itself generated within the cells.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Paraffin rat retinal sections.

Primary Incubation

1/1000 dilution in 3% BSA, overnight at four degrees on shaker.

Blocking Agent

5% BSA/1 hr at RT

Secondary Incubation

3%PBS/1 hr at RT

Tertiary Incubation

None

Detection

Chemiluminescence

Results Summary

Observed rhodopsin expression in photoreceptor layer of various rat retinal sections. Dilution: 1:1000 (IF).

Additional Notes

Take the 4µ paraffin retina sections then transfer into xylene and gradient alcohols (90, 70, and 60%). Next add proteinase-K/10min. Wash with PBS. Block the sections with 5% PBS/1 hr. After washing, go for incubation with this Ab in 3%PBS overnight/4oC. Next day wash and incubate with 2OAb in 3%PBS/1 hr. Then wash and mount with DAPI. Check in microscope.
