The quantification of oxidative burst of peripheral blood mononuclear cells (PMN) was performed to compare respiratory burst function in tumor-bearing dogs with healthy dogs.

Heparinized blood were incubated with opsonized E.coli or control solution for 10min at 37°C in a water bath. Samples were then incubated with dihydrorhodamine 123 as a fluorogenic substrate for respiratory burst at RT in the dark for 20 minutes. This reaction was extinguished, erythrocytes were removed, and cells were fixed by the addition of lysing solution and incubation for 10 min at RT. DNA was stained by DNA staining solution (PI) and the percentage of cells performing oxidative burst and MFI of positive cells was measured using flow cytometry.
Erythrocytes need to be removed completely to have better results.
A significant decrease in the generation of respiratory burst in PMNs from dogs with cancer compared to healthy dogs.
Samples better tested within 30 min using flow cytomety.
