Our lab works with kinases involved in kidney cancer, including STAT3. We needed to reliably detect low protein levels of phosphorylated STAT3 (at Tyr705) in 786-0 cell lysates, and had no problem doing so with this antibody. We always start out with Cell Signaling antibodies because of their reputation, and we're glad once again that we did.
Western Blot

1:1000 Dilution Overnight in 5% BSA in 1X TBS-T at 4C
5% Milk in 1X TBS-T
1:5000 dilution of HRP-conjugated Rabbit secondary in 5% Milk in TBS-T for 1 hour at RT

ECL/FEMTO --> Autoradiography Film via HRP-Conjugate Rabbit Secondary Antibody
With no need for additional optimization, we were able to accurately and reliably detect phospho-STAT3 levels in our cell lysates. We went down to 20 ugrams per well and were still able to detect the protein, which was particularly impressive. Great results.
