I have used SensiFAST SYBR Lo-ROX master mix for qPCR experiments to measure bacterial gene expressions. I have also compared the gene expression results with Agilent's Brilliant III Ultra Fast qPCR master mix. Gene expression as well as standard curves obtained with SensiFast kit were comparable with Agilent's kit. SeniFast being slightly less expensive than Agilent's will be the choice to go with for qPCR experiments in my lab.

Starting Material

E. coli cDNA

Protocol Overview

cDNA were diluted to 1:10 in nuclease free water. 10 fold diluted DNA standards (6 points dilutions) were prepared. Then 10 microliter of 2X sensifast master mix along with 500 nm of each forward and reverse primers were used in a 20 microliter reaction. PCR cycling conditions were followed as mentioned in instruction manual of the kit.

Tips

Protect the SensiFAST kit from exposure of light.

Results Summary

Comparable results of gene expression within dynamic range of detection (as extrapolated from standard curve) and also comparable with Agilent's ultra fast sybr green master mix.
