Mice were injected IP with heat-killed E. coli and peritoneal lavage recovered. Immune populations were identified by flow cytometry on total peritoneal lavage. 7-AAD was added to exclude dead cells.

Peritoneal cells - 8h after heat-killed E. coli injection
Cells were incubated 1:200 Ly6c-eFlour450 1:400 Ly6g-APC/Cy7 - 30 minutes on ice, blocked with 1:100 FcBlock for 5 minutes on ice, and then incubated 1:80 with 7AAD on ice. They were then imaged with LSRII Flow Cytometer.

Dead cells were easily excluded by 7AAD staining of cells with compromised cell membranes.

Inexpensive, bright staining
Inexpensive live/dead stain allows exclusion of dead cells from flow analysis. 500 tests is $35 at the time of writing.
