Thymocytes were stained for in vivo injection and tracking, this experiment was to test the intensity of staining and select an optimal staining condition.

Cells were incubated in 5uM CellTracker Green in PBS @ 37*C for 20 minutes. Reaction was quenched with media containing FBS. Cells were collected in FITC channel on BD LSR II.

Cells stained brightly with CellTracker Green and were easily identified after recover from injection IP.
Staining is time and concentration dependent so it's very easy to optimize.
A great cell dye in the FITC channel.
