I am interested in the expression of urinary microRNAs as potential biomarkers for haematological diseases. I have used the Noregn Biotek Urine microRNA Purification Kit to successfully isolate microRNA from as little as 1ml of urine. The purified RNA was quantified on a Bioanalyzer and used for downstream qRT-PCR using TaqMan MicroRNA Assays.

Fresh frozen urine (up tp 5ml).
The protocol is very straightforward and quick to perform, and can be applied using 1-15ml of starting material. Briefly, lysis buffer is initially added (with or without additional B-mercaptoethanol) and vortexed to lyse cells. Ethanol is then added, and the sample centrifuged through a kit-supplied spin column. When all material has been passed through, the column is then washed using the supplied wash buffer, dried and the kit elution solution applied. The final RNA is eluted in 50ul volume and can be used for downstream analysis.
A number of samples can be processed at one time
microRNA was successfully recovered from small volumes. RNA could be quantified (Bioanalyzer) and used for downstream qRT-PCR (TaqMan Assays).

Overall, this is a good kit for the isolation of microRNA from relatively small volumes of urine. Isolated RNA is suitable for downstream qRT-PCR.
