In order to find out what cells play a roll in the proteoglycan (PG)-induced arthritis (PGIA) model, we identify the cell populations in the different tissue of PGIA mice by flow cyotemetry. In this experiment, 1X10^6 splenocytes or peritoneal cells treated with PMA(25ng/ml) and Ionomycin (500ng/ml) at 37oC for 4 hours. Then cells are harvested and stained with 0.5ul PE-Cy anti-mouse CCR4 and other Abs in 50ul FACS buffer for 30 min at 4oC.
