The BD FACSCalibur allows users to perform cell analysis for a wide range of applications including enumeration of lymphocyte subsets, stem cells, residual white blood cells, and reticulocytes. It is a 2 laser, 4 color system. It has a 488nm blue laser and 637nm red laser, with 4 filters- 525BP, 585 BP, 670 LP, 650 LP. In our lab, this instrument is primarily used for diagnostic samples.
Flow cytometry
Cells in suspension.
Cells are usually treated with antibody of choice and the end product is brought into suspension with PBS and analyzed.
Making sure the cells are not too big or clumpy or it will clog the machine.
It is able to detect up to 4 colors. We used it in Detection of Bcell, T cell lymphoma, Red blood cells bound to antibody, etc.
Very user friendly and is a work horse.
