The BD LSR Fortessa X-20 Cell Analyzer is a great instrument for looking at multiple cell markers within a sample. Only one sample is needed for up to 18 colors. The ability to look at multiple markers allows for better profiling of samples. This is especially useful for in vivo studies. In our studies, we were able to look at T cells, B cells, Monocytes, Macrophages, Neutrophils, and Dendritic cells, all within one sample.

Protocol Overview

Single cell suspensions were prepared from mouse spleen and fluorescently labeled with various cell surface markers within one sample. The LSR Fortessa was used to detect the cell markers.

Tips

Compensation can be done on the Diva software the Fortessa is linked to or compensation can be done on Flow Jo if that is the software analysis that is being used.

Results Summary

We were able to look at a variety of markers without setting up multiple samples. B cells, T cells, Monocytes/Macrophages, and Dendric Cells can all be analyzed simultaneously.

The Bottom Line

The BD LSR Fortessa X-20 is a very useful instrument to have in the lab. It has a lot of quirks and takes a few tries for new users to be comfortable using it. However, with the ability to detect multi-color parameters all within one sample, a lot of time is saved during setup. Users can better profile their samples quickly and efficiently. This instrument is highly recommended.
