In contrast to compensating for fluorophore antibodies, to compensate for the live/dead stains, amine linked beads are required. This further allows differentiation of live or dead cells from staining as opposed to the cell's morphology.

Protocol Overview

These beads are only used for one well, to compensate for the live/dead fixable stain. In a round bottomed well, 2 drops of the positive and 2 drops of the negative beads are added. These are then spun down and resuspended in 50ul of MACS buffer, before 0.5ul of live/dead stain is added. Incubate these beads for at least 15 mins at 4C in the dark, before washing the beads twice with MACS buffer. Fix the beads in 2% formaldehyde prior to running on the Flow Machine.

Tips

These beads are only for amine-linked dyes, such as the live dead stain, not for individual antibody conjugates.

Results Summary

These beads successfully allow the differentiation between live/dead stained beads (cells) or not.
