I am using the NB600-633 antibody to stain for cytokines following microglia M1 suppression using Minocycline 30mg/kg in rat brain paraffin tissue. This antibody was used to stain IL-1B in rat brain section.

Experimental Design and Results Summary

Applications

Immunohistochemistry

Sample

4%paraformaldehyde fixed rat brain; 6 um paraffin sections

Primary Incubation

Incubate in Primary overnight at 4oC (1:300 dil of NB600-633 in PBS containing 1%NGS and 0.3%Triton x)

Blocking Agent

5%NGS with PBS containing 0.3% Triton x for 1 hour

Secondary Incubation

Vector lab pk4000 secondary biotinylated antibody (1/2000) in PBST with 2%NGS at RT for 1 hour. Meanwhile make up ABC reagent according to kit instruction in PBST.

Tertiary Incubation

ABC reagent 1 hour at RT.

Detection

DAB and H2O2

Results Summary

Get staining that looks like cells under 200x magnification

Additional Notes

Got specs which look like cytokine staining. Citrate unmasking is necessary to enhance staining.
